
Soooo.....I'll tell you the moral of the story ahead of time okay? Here goes: Never trust what is written in a journal article methods w/out emailing the authors directly!!! See...I broke down and wrote the scientist that gave us the antibody that I have been unable to get to work. Told him my protocol and asked him if I was missing something from his paper.....turns out I was...what wasn't written!!! Moreover, what was written was wrong. I know because I spent and hour today double checking. The one paper he describes the assay I'm doing, it clearly states that the antibody is to be used platebound to stimulate cells. Now he tells me to use it soluble...moreover, that using it platebound will cause it to be inhibitory.

Gorramit. This is why we don't have a cure for cancer people!! Seriously. It was all I could do today to not hit computer when I read it (i was at work, so my baby wasn't at risk). Auuuuggghhh! I wasted a year on this because I assumed i was doing something wrong when it turns out that he or his postdoc fucked up when they wrote the goddamn paper!! Tianna.
And i apologize for the language, but trust me, Simon would be saying the exact same things.
On the other hand, it looks like our plasmid finally got made right last week after all (i just didn't know what it was supposed to look like).
So yeah...things are mildly better and more encouraging. So keep sending me your luck, obvouisly I have none of my own.
